Non-competitive Inhibition


Non-competitive inhibitors bind non-covalently to sites other than the substrate binding site.

Therefore, the binding of the substrate and the inhibitor are independent of each other and the binding of an inhibitor cannot be reversed by increased [S]. They alter the Vmax but not the Km. The Ki for noncompetitive inhibition is much more complex and involves EI and ESI.
